Animal Model of Renal Failure

Chronic renal failure (CRF) refers to chronic kidney disease caused by decreased glomerular filtration rate and associated with metabolic disorders and clinical symptoms of the syndrome. The use of animal models can help to understand the occurrence and development of CRF and the prevention and control measures. In recent years, with the deepening of experimental research, CRF experimental animal model modeling methods have more experience and further development.


Nephrectomy model has a lot of ways to do.  Following is the Physical Method.



Nephrectomy model has a long history, as early as 1952 Platt and other rats with 5/6 nephrectomy successfully produced CRF animal model. China's Sha Zhaohui and other reports of rat 5/6 nephrectomy using two operations: thigh intramuscular injection of anesthetized rats, select the back of the left incision, exposure to the left kidney, stripping kidney capsule, the upper and lower kidney of the 1/3 resection, gelatin sponge oppression hemostasis.


7 days after surgery, the second phase of surgery, removal of the right kidney. 90 days after surgery stable in azotemia. Wang Ruining and his team remove arc resection 2/3 of the renal tissue, the main removal of the cortex. 1 week after the removal of the entire right kidney, 2 times a total of 80% of the removal of the kidneys, 8 weeks after the success of modeling.


5/6 nephrectomy CRF model is consistent with the glomerular hyper perfusion, high filtration, high pressure caused by renal failure theory, this model to glomerular hypertrophy, hardening as the main features, showing the same process with human kidney fibrosis, so the model performance close to the clinical study to reduce renal perfusion, high filtration and inhibition of meningeal cell proliferation of drugs can be used in this model.


In this study, a series of changes in renal bone disease such as low calcium and high phosphorus were observed in the 8th week of the experiment, suggesting that this model can also be used as a research model of renal bone disease. But the model needs to do two surgeries, surgical operation is complex, need to master the removal of renal tissue technology, should not cut to the renal pelvis, so as to avoid leakage of urine.


And the modeling method is easy to cause bleeding, infection and even death; the model preparation time is long. In addition, there are 3/4,6/7,7/10 and other nephrectomy, different models of biochemical indicators and pathological damage were manifested as chronic renal failure, but the model is not as stable as the 5/6 nephrectomy model.
